  • GluA2-lacking Ca2+-permeable AMPARs (CP-AMPARs) play integral roles in synaptic plasticity and can mediate excitotoxic cellular signaling at glutamatergic synapses. However, the developmental profile of functional CP-AMPARs at the auditory brainstem remains poorly understood. Through a combination of electrophysiological and live-cell Ca2+ imaging from mice of either sex, we show that the synaptic release of glutamate from the calyx of Held nerve terminal activates CP-AMPARs in the principal cells of the medial nucleus of the trapezoid body in the brainstem. This leads to significant Ca2+ influx through these receptors before the onset of hearing at postnatal day 12 (P12). Using a selective open channel blocker of CP-AMPARs, IEM-1460, we estimate that ∼80% of the AMPAR population are permeable to Ca2+ at immature P4–P5 synapses. However, after the onset of hearing, Ca2+ influx through these receptors was greatly reduced. We estimate that CP-AMPARs comprise approximately 40% and 33% of the AMPAR population at P18–P22 and P30–P34, respectively. By quantifying the rate of EPSC block by IEM-1460, we found an increased heterogeneity in glutamate release probability for adult-like calyces (P30–P34). Using tetraethylammonium (TEA), a presynaptic potassium channel blocker, we show that the apparent reduction of CP-AMPARs in more mature synapses is not a consequence of presynaptic action potential (AP) speeding.   • My I.S. project studies several Chinese films that are elaborations of the Chinese Communist Revolution. They include White Haired Girl by A Jia and Zheng Yiqiu (1950), Red Detachment of Women by Xie Jin (1961), Sparkling Red Star by Li Ang and Li Jin (1974), Breaking With Old Ideas by Li Wenhua (1975), To Live by Zhang Yimou (1993), and Through Stunning Storms by An Zhanjun (2011). The study of these films on Chinese revolution also incorporates my reflections the Long March enactments organized through China Studies Institute at Peking University, and my interviews with Chinese individuals, made possible by a grant from Copeland Funds for undergraduate researches. My reflections on the Long March trip, completed during the spring of 2013, provide a sense of reality and history for such famous battle sites in Guizhou Province as Loushanguan, Chishui River, Qinggangshan, and Zhunyi where the Red Army fought. Along with a few photos I took, these reflections help capture the monumental struggle of Chinese Communism that film directors hope to convey. My analyses of the films and the interviews conducted in Beijing with Chinese people varying in age and social class show paralleling and sometimes conflicting narratives of history and film to record the key events in modern Chinese history. Most importantly, interviews on people’s reactions to these films and inevitably their understanding of the Communist Revolution reveal drastic change and difference in Chinese politics. This...

  • Root rot of Paris polyphylla has received widespread attention due to its threat to yield and leads to serious economic losses. However, the relationship among the rhizosphere microbial community, metabolites and root rot disease remained largely unexplored. Herein, we used integrated 16S rRNA, ITS, RNA sequencing and UPLC-MS/MS to systematically investigate the differences between healthy and diseased P. polyphylla. We found that root rot reduced the microbial diversity in the diseased P. polyphylla compared with the healthy control. The relative abundance of the bacterial phylum Actinobacteria increased in the diseased rhizome of P. polyphylla. For the fungal community, root rot disease contributed to an increased relative abundance of Ascomycota and decreased Glomeromycota at the phylum level. The transcriptomic results showed that the differently expressed genes were significantly enriched in the “Biosynthesis of various alkaloids”, “flavonoid biosynthesis” and “isoflavonoid biosynthesis” and “Phenylpropanoid biosynthesis” was dramatically enriched in healthy P. polyphylla compared with that in diseased P. polyphylla. Likewise, the metabolomic results showed that the biosynthesis of secondary metabolites and metabolic pathways was found to be significantly enriched by differential metabolites.
